Abstract

The current study was aimed to isolate and identify Pseudomonas putida from Harir soil and then evaluating their phosphate solubilizing efficiency and screening for the presence of phosphate solubilizing genes. Soil samples were collected from different rhizosphere plants grown in Harir area- north of Erbil governorate. A total of 25 isolates of Pseudomonas sp. were isolated from different soil samples, eleven fluorescent pseudomonad isolates were identified as Pseudomonas putida using morphological features, cultural, biochemical characteristics and 16S rRNA gene sequencing technique. These isolates resolute for their phosphate solubilizing efficiency and all of them showed phosphate solubilizing activity. Isolates (Pp6 and Pp8) were the most efficient isolates, while (Pp2 and Pp5) were the least efficient isolates in P-solubilization. Pp6, Pp8, Pp2, and Pp5 were selected to screen for the presence of phosphate solubilizing genes (namely phosphonoacetase, phosphohydrolase, and phytase genes), using specific primers with PCR technique. The results showed positive reactions and all isolates contained phosphate solubilizing genes, and phylogenetic analysis showed that these isolates PP6, PP8, PP2, PP5 were found to be most similar to Pseudomonas putida S16 (NCBI GenBank Acc . No:Assembly: GCA_000219705.1) Bio Project:67881 and a maximum of 97% identity was reputable.
